目的探讨芍药苷对过氧化氢(H2O2)诱导SH-SY5Y氧化损伤的保护作用及其可能机制。方法MTT法测定细胞存活率,细胞免疫化学技术检测细胞细胞核增殖抗原(PCNA)、Caspase-3的表达。结果 0.87mmol/L H2O2可使细胞存活率降低,PCNA的表达减少,Caspase-3的表达增加。经过39μg/mL和62.5μg/mL浓度的芍药苷处理后,H2O2诱导的SH-SY5Y细胞存活率明显升高,PCNA的表达明显增加,同时抑制Caspase-3的表达。结论芍药苷可抑制过氧化氢诱导的SH-SY5Y细胞凋亡,其作用机制可能与其增加细胞PCNA的表达,减少Caspase-3的表达有关。
Objective To investigate the protective effect of paeoniflorin on oxidative damage of SH-SY5Y induced by hydrogen peroxide (H2O2) and its possible mechanism. Methods The cell viability was determined by MTT assay. The expression of PCNA and Caspase-3 was detected by immunocytochemistry. Results After treated with 0.87 mmol / L H2O2, the cell viability decreased, the expression of PCNA decreased and the expression of Caspase-3 increased. After 39μg / mL and 62.5μg / mL paeoniflorin treatment, H2O2-induced SH-SY5Y cell survival rate was significantly increased, PCNA expression was significantly increased, while inhibiting Caspase-3 expression. Conclusion Paeoniflorin can inhibit hydrogen peroxide-induced apoptosis of SH-SY5Y cells, and its mechanism may be related to increasing the expression of PCNA and decreasing the expression of Caspase-3.
