采用微管吸吮实验技术并结合十二烷基硫酸钠聚丙烯酰胺凝胶电泳( S D S P A G E) 等生化方法研究遗传性球形红细胞增多症( H S) 红细胞膜粘弹特性及膜骨架蛋白的变化。结果表明: H S红细胞膜的弹性模量和正常对照组比较无明显差异而粘性系数显著高于正常值,细胞粘滞性增大,变形能力降低;在红细胞膜骨架蛋白中,收缩蛋白、带3 蛋白和带5 蛋白的含量低于正常细胞,膜骨架蛋白的巯基含量低于正常细胞。这一结果提示在膜骨架蛋白网络中,其结构和成分的完整性在决定红细胞变形行为上起着重要作用。
The viscoelastic properties of hereditary spherocytosis (Hs) erythrocyte membrane were studied by using microtubule sucking technique combined with sodium dodecyl sulfate-polyacrylamide gel electrophoresis (S D S-P A G E) And membrane scaffold protein changes. The results showed that the elastic modulus of erythrocyte membrane was not significantly different from that of the normal control group, while the viscosity coefficient was significantly higher than that of the normal control group. The cell viscosity increased and the deformability decreased. In the erythrocyte membrane scaffold protein, 3 protein and band 5 protein content is lower than normal cells, membrane scaffold protein thiol content is lower than normal cells. This result suggests that the structure and compositional integrity of the membrane scaffold protein network play an important role in determining erythrocyte deformability.
