目的研究青蒿素哌喹片(ATQ)对昆明种小鼠灌胃给药的急性毒性。方法按Bliss法设计试验,对小鼠单次灌胃ATQ,测定ATQ对小鼠半数致死量(LD50)和LD50的95%可信限,观察14 d内小鼠的毒性反应和死亡情况,对小鼠进行大体解剖,肉眼观察并进行组织病理学检查。结果 ATQ单次灌胃给药可使小鼠出现不同程度的毒性反应,包括自发活动减少、闭眼、俯卧、运动失调、震颤、惊跳、强制性-阵挛性抽搐、角弓反张。小鼠LD50为2802.38 mg·kg-1,LD50的95%可信限为2343.51~3520.11 mg·kg-1,死亡小鼠肉眼观察未发现各脏器异常。结论 ATQ对小鼠单次灌胃给药具有一定的毒性,涉及的毒性靶器官可能是中枢神经系统。
Objective To study the acute toxicity of artemisinin and piperaquine tablet (ATQ) administered orally to Kunming mice. Methods According to the Bliss design test, mice were given a single oral gavage of ATQ. The 95% confidence limits of LD50 and LD50 of ATQ against mice were determined. The toxicity and mortality of mice were observed within 14 days. Mice were grossly dissected, macroscopically and histopathologically examined. Results A single intragastric administration of ATQ resulted in different degrees of toxic reaction in mice, including decreased spontaneous activity, eyes closed, prone position, dyskinesia, tremor, asthma, forced-clonic convulsion and cornering bow reversed. The LD50 of mice was 2802.38 mg · kg-1, and the 95% confidence interval of LD50 was 2343.51 ~ 3520.11 mg · kg-1. No abnormalities of organs were found in the dead mice. Conclusion ATQ has some toxicity to single oral administration to mice, and the toxic target organ involved may be the central nervous system.
