目的探讨FTY720、环孢菌素A(Cs A)及两药联合应用对弗氏完全佐剂诱导性大鼠关节炎模型的治疗效果。方法 60只大鼠按照随机数字表法分为5组:正常对照组、模型对照组、FTY720治疗组、Cs A治疗组及FTY720+Cs A治疗组,每组12只。ELISA法定量检测关节液中TNF-α和IL-1β含量;28 d时处死大鼠取大鼠胸腺、脾脏,称重后评价胸腺指数及脾脏指数。结果 (1)HE染色结果:FTY720治疗组和Cs A治疗组与模型对照组相比,可见血管翳形成,但管腔略小,未见炎细胞形成;FTY720联合Cs A组被覆滑膜细胞下可见纤维组织增生,未见血管翳形成。(2)胸腺指数、脾脏指数及TNF-α、IL-1β含量:FTY720治疗组和Cs A治疗组较正常对照组升高,且差异有统计学意义(P<0.05),与模型对照组相比含量降低;FTY720联合Cs A组与正常对照组相比,差异无统计学意义(P>0.05),与模型对照组相比,差异均有统计学意义(P<0.05)。结论 FTY720联合Cs A可通过降低大鼠RA模型的胸腺指数、脾脏指数及TNF-α、IL-1β在类风湿性关节炎中起到治疗作用,但两者单独应用效果略差。
Objective To investigate the therapeutic effect of FTY720, cyclosporine A (Cs A) and their combination on the complete adjuvant-induced arthritis model in rats. Methods Sixty rats were randomly divided into five groups according to random number table: normal control group, model control group, FTY720 treatment group, CsA treatment group and FTY720 + CsA treatment group, 12 in each group. ELISA method for quantitative determination of TNF-α and IL-1β in synovial fluid; rats were sacrificed 28 days after the thymus and spleen of rats were weighed, and the thymus index and spleen index were evaluated after weighing. Results (1) The results of HE staining: compared with the model control group, FTY720 treatment group and CsA treatment group showed the formation of pannus, but the lumen was slightly smaller and there was no inflammatory cell formation; FTY720 and CsA group were coated with synovial cells Visible fibrous tissue proliferation, no formation of blood vessels 翳. (2) Thymus index, spleen index, TNF-α and IL-1β levels: FTY720 treatment group and CsA treatment group than the normal control group increased, and the difference was statistically significant (P <0.05) (P <0.05). Compared with the model control group, the difference was statistically significant (P <0.05) .Conclusion: FTY720 combined with CsA group has no significant difference compared with the normal control group (P> 0.05). Conclusion FTY720 combined with CsA can reduce the thymus index, spleen index, TNF-α and IL-1β in rat RA model and play a therapeutic role in rheumatoid arthritis. However, the effect of FTY720 and Cs A alone is slightly worse.
