目的以乌骨鸡为原料,优选生物酶酶解乌骨鸡的工艺条件。方法以光密度法测定酶解液氨基酸为指标,对酶解温度、时间、pH和酶底比进行比较,用正交试验设计法筛选酶解最佳工艺条件。以乌鸡白凤丸补气养血、调经止带主要药效为指标,对酶解提取工艺进行验证。结果确定酶解方案为先用pH8.3的人工肠液湿润,90℃加热,降温后转入50℃恒温水浴,加入12000U中性蛋白酶酶解4h。结论以乌骨鸡酶解工艺入药的乌鸡白凤丸不仅保持了原制剂工艺的作用,酶解工艺对测试的各项指标均有明显的改善作用,显示出良好的效果。
Objective To optimize the technological conditions for enzymatic hydrolysis of black bone chickens using black bone chicken as raw material. Methods The optical density method was used to determine the amino acid in the hydrolysate. The temperature, time, pH and enzyme base ratio were compared. The orthogonal experimental design method was used to screen the optimum conditions of enzymolysis. Taking Wuji Baifeng Pills as the index of qi and nourishing blood and regulating menstruation, the enzymatic extraction process was verified. The results confirmed that the enzymolysis solution was first moistened with artificial intestinal fluid with pH 8.3, heated at 90°C, cooled, transferred to a constant temperature water bath at 50°C, and added with 12000 U of neutral protease for 4 hours. Conclusion The Wuji Baifeng Pills, which were treated with Wuji Baifeng Pills in the enzymolysis process of black-bone chickens, not only maintained the original preparation process, but also had obvious improvement effects on the tested indicators and showed good results.
