目的:采用HPLC法测定盐酸芬氟拉明血药浓度,对5只家兔进行药代动力学研究,并就一例芬氟拉明中毒致死者的血、尿和脏器检材的毒物检测进行了分析。方法:流动相为甲醇-002mol·L-1庚烷磺酸钠(25∶75),二苯胺为内标,在268nm进行检测。结果:芬氟拉明线性范围为5~200μg·L-1(r=09986),最低检测浓度5ng,平均方法回收率为977%。日内、日间RSD均小于6%,达峰时间为(15±3)h,峰浓度(654±49)μg·L-1,药时曲线下面积为(1754±328)μg·L-1·h-1。结论:此方法稳定、可靠,可用于血药浓度分析和法医中毒检测。
OBJECTIVE: To determine the concentration of fenfluramine hydrochloride by HPLC and to study the pharmacokinetics of five rabbits, and to detect the poisonous substances in blood, urine and organ samples of one death caused by fenfluramine poisoning Analysis. Methods: The mobile phase consisted of methanol-0.02 mol·L-1 heptanesulfonic acid sodium (25:75), diphenylamine as internal standard and detection at 268 nm. Results: Fenfluramine linear range of 5 ~ 200μg · L-1 (r = 09986), the lowest detection concentration of 5ng, the average recovery rate was 977%. The intra-day and inter-day RSD were less than 6%, peak time was (15 ± 3) h, peak concentration (654 ± 49) μg · L-1, ± 328) μg · L-1 · h-1. Conclusion: This method is stable and reliable and can be used for the analysis of blood concentration and forensic poisoning.
