自放射免疫检测法(RIA)用于检测胰岛素以来,胰岛素的实验性研究和临床检测有了很大的进展。但RIA长期大量应用,其放射性同位素不仅对检测者自身具有危害,且对自然环境亦造成污染。70年代初,Engvall建立酶联免疫吸附试验(Enzyme Linked ImmunosorbentAssay,ELISA)用于检测体内微量抗原与抗体,Miedema将ELISA法应用于检测胰岛素。但目前ELISA检测胰岛素的方法,尚存在操作繁琐,流程费时或试剂不易得到等问题。我们以辣根过氧化物酶标记胰岛素抗血清,采用ELISA夹心法,用聚苯乙烯微板的小孔作为载体,进行检测胰岛素的研究,并与RIA法对比,取得较为满意的结果。现报告如下。
Since the radioimmunoassay (RIA) for the detection of insulin, experimental research and clinical testing of insulin have made great progress. However, RIA has been used in large quantities for a long time. Radioisotopes of the RIA not only endanger the testers themselves but also pollute the natural environment. In the early 1970s, Engvall established Enzyme Linked Immunosorbent Assay (ELISA) to detect trace antigens and antibodies in vivo, and Miedema applied ELISA to detect insulin. However, the current ELISA detection of insulin methods, there are still cumbersome operation, the process time-consuming or difficult to obtain reagents and other issues. Insulin antiserum was labeled with horseradish peroxidase (HRP). ELISA sandwich method and micropore of polystyrene microplate were used as carriers to detect insulin. Compared with RIA method, we obtained satisfactory results. The report is as follows.
