目的:研究瓜蒌皮对高血脂合并心肌缺血(HL-AMI)大鼠CD_(34)~+细胞的动员作用,以及对金属基质蛋白酶9(MMP-9)蛋白表达量和酶活性的调节作用,从促进干细胞动员的角度揭示该药材干预冠心病痰浊壅塞证的机制。方法:雄性Wistar大鼠随机分为对照组、模型组、瓜蒌皮干预组,每组再分为7个亚组。各亚组大鼠分别在急性心肌缺血(AMI)后第1-7天处死,腹主动脉取血及心脏,检测外周血CD_(34)~+细胞含量及血浆、心肌组织中MMP-9含量。结果:AMI后,模型组大鼠CD_(34)~+细胞含量于第6天达到峰值并开始下降(P<0.05),MMP-9活性及表达量第3天达到峰值,第4天即下降到基线水平;瓜蒌皮干预组大鼠CD_(34)~+细胞含量7d内一直上升,且上升幅度明显高于模型组(P<0.05),MMP-9活性及表达量于第5天达到峰值,且一直明显高于同时期的模型组(P<0.05)。结论:瓜蒌皮可显著促进HL-AMI大鼠骨髓干细胞的动员,其机制可能与上调MMP-9活性及蛋白表达量有关。
OBJECTIVE: To study the mobilization effect of melon skin on CD34 (34) + cells in hyperlipidemic rats with myocardial ischemia (HL-AMI) and the regulation of MMP-9 protein expression and enzyme activity From the perspective of promoting the mobilization of stem cells, we can reveal the mechanism of the herbal medicine in treating phlegm and blood stasis syndrome of coronary heart disease. Methods: Male Wistar rats were randomly divided into control group, model group and control group. Each group was divided into 7 subgroups. Rats in each subgroup were sacrificed on days 1-7 after acute myocardial ischemia (AMI), blood and heart were taken from the abdominal aorta, and the levels of CD34 + cells in peripheral blood and the levels of MMP-9 in plasma and myocardium content. Results: After AMI, the content of CD_ (34) ~ + cells in model group peaked on the 6th day and began to decrease (P <0.05). The activity and expression of MMP-9 peaked on the 3rd day and decreased on the 4th day (P <0.05). The activity and expression of MMP-9 reached its peak on the 5th day in the model group (P <0.05) Peak, and has been significantly higher than the same period model group (P <0.05). Conclusion: Gulupi can significantly promote the mobilization of bone marrow stem cells in HL-AMI rats, which may be related to the up-regulation of MMP-9 activity and protein expression.
