以超速离心法制备大鼠肝刺激因子(HSS)。采用无血清培养技术培养大鼠肝细胞并观察HSS作用肝细胞后不同时刻肝细胞EGF受体mRNA表达的变化。结果显示,HSS可刺激EGF受体mRNA的表达。给药后2~6h,mRNA表达达高峰。HSS与EGF合用可明显刺激受体mRNA表达。Northern杂交分析表明,mRNA表达明显增高系由于5.6和10.0kb两基因片段所致,提示HSS可在基因水平调节EGF受体活性。本工作进一步证明HSS对EGF受体的调节可能为其促肝再生的重要机制之一。
Rat hepatic stimulating factor (HSS) was prepared by ultracentrifugation. Serum-free culture of rat hepatocytes was used to observe the changes of EGFR mRNA expression in hepatocytes at different time points after HSS treatment. The results show that HSS can stimulate the expression of EGF receptor mRNA. After 2 ~ 6h after administration, mRNA expression peaked. HSS combined with EGF can obviously stimulate the receptor mRNA expression. Northern blot analysis showed that mRNA expression was significantly increased due to the 5.6 and 10.0 kb two gene fragments, suggesting that HSS can regulate EGF receptor activity at the gene level. This work further demonstrated that HSS EGF receptor regulation may be one of the important mechanisms of promoting liver regeneration.
