目的:研究膜型基质金属蛋白酶1(membranetype1matrixmetalloproteinase,MT1MMP)基因mRNA在人胃癌组织中的表达及其与胃癌浸润转移和临床分期的关系。方法:应用RTPCR半定量的方法检测MT1MMP基因mRNA在30例胃癌组织、正常粘膜和转移淋巴结中的表达。使用SPSS/PC+统计软件进行t检验和多因素分析。结果:MT1MMP基因的mRNA在肿瘤组织和转移淋巴结中分别有76.7%(23/30)和76.9%(10/13),高于正常组织。按TNM分期,MT1MMP在Ⅰ、Ⅱ期与Ⅲ、Ⅳ期的表达差异有显著性。经多元回归分析,MT1MMP基因的表达与肿瘤的浸润深度、有无淋巴结转移及有无癌栓正相关。结论:MT1MMP基因参与胃癌的直接浸润和转移,可能成为一种潜在的肿瘤生物学标记物。
Objective: To investigate the expression of membrane type 1 matrix metalloproteinase 1 (MT1 MMP) mRNA in human gastric cancer and its relationship with invasion, metastasis and clinical staging of gastric cancer. METHODS: Semiquantitative RTPCR was used to detect the mRNA expression of MT1MMP gene in 30 cases of gastric cancer, normal mucosa and metastatic lymph nodes. The t-test and multivariate analysis were performed using SPSS/PC+ statistical software. RESULTS: The mRNA of MT1 MMP gene was 76.7% (23/30) and 76.9% (10/13) respectively in tumor tissues and metastatic lymph nodes, which was higher than normal tissues. According to TNM staging, there was a significant difference in the expression of MT1 MMP in stage I, II and III and IV. By multiple regression analysis, the expression of MT1MMP gene was positively correlated with the depth of tumor invasion, presence of lymph node metastasis, and presence or absence of cancer thrombus. Conclusion: MT1 MMP gene is involved in the direct invasion and metastasis of gastric cancer, which may become a potential tumor biomarker.
