传统的无细胞百日咳疫苗生产工艺包括大发酵罐培养细菌、硫酸铵沉淀、解毒、高盐浓度抽取、透析和蔗糖梯度超离心等步骤。在生产过程中去除了百日咳菌体中的有毒成分,提取纯化出有效抗原,使无细胞百日咳疫苗在保持免疫原性的基础上大大减少了副反应的发生。在抗原的提取过程中,可以对不同抗原组分分别纯化后按一定比例混合,也可以对某些组分联合提取。
