目的初步探索抗体免疫脂质磁球法在肺癌循环肿瘤细胞(CTCs)检测中的应用。方法采用反向蒸发法制备上皮细胞粘附分子(Ep CAM)和表皮生长因子受体(EGFR)抗体免疫脂质磁球。在磷酸盐缓冲溶液(PBS)和外周血肺癌CTCs模型中,评价Ep CAM和EGFR抗体免疫脂质磁球对A549细胞的捕获效率;在此基础上,用上述两种抗体免疫脂质磁球法检测肺癌患者外周血中CTCs;并将Ep CAM免疫脂质磁球法与CellsearchTM系统进行比较。结果 Ep CAM和EGFR免疫脂质磁球法在PBS溶液CTCs模型中对A549细胞的中位检出率分别为92%和90%,在外周血模型中分别为85%和81%;两种磁球法在13例肺癌患者中均检测出了CTCs,中位检出个数均为5个,而在健康对照者中中位检出个数均为0个(P<0.05)。用Ep CAM免疫脂质磁球法和CellsearchTM系统检测3例肺癌患者的CTCs,前者对其中2例患者的CTCs检出量高于后者,另外1例相等。结论 Ep CAM和EGFR免疫脂质磁球法均能有效检出肺癌CTCs,两种磁球法检出率相当。Ep CAM免疫脂质磁球法对肺癌患者CTCs检出量有高于CellsearchTM系统的趋势。
Objective To explore the application of antibody-immunodominant lipid magnetic ball method in the detection of circulating tumor cells (CTCs) in lung cancer. Methods EpCAM and epidermal growth factor receptor (EGFR) antibody immunoliposomes were prepared by reverse evaporation method. In the phosphate buffered saline (PBS) and CTCs model of peripheral blood lung cancer, the capture efficiency of EpCAM and EGFR antibody-immunized lipid magnetic sphere on A549 cells was evaluated. On the basis of this, the immunoglobulin magnetic beads method To detect CTCs in peripheral blood of patients with lung cancer; and compare the Ep CAM immunoliposomal magnetic sphere method with CellsearchTM system. Results The median detection rates of Ep CAM and EGFR immunostimulosphere were 92% and 90% in A549 cells in PBS solution CTCs model respectively, and 85% and 81% in peripheral blood cells respectively. CTCs were detected in all 13 lung cancer patients by ball method. The median number of detected CTCs was 5 in all lung cancer patients and 0 in healthy controls (P <0.05). EpCAM immunocell magnetic ball method and CellsearchTM system were used to detect CTCs in 3 patients with lung cancer. The former had higher detection rate of CTCs in 2 patients and the other 1 patient was equal. Conclusions Both Ep CAM and EGFR immunoliposomal magnetic ball methods can effectively detect CTCs in lung cancer. The detection rates of the two kinds of magnetic ball methods are quite high. EpCAM immunoglobulin magnetic ball detection of lung cancer patients with CTCs higher than the CellsearchTM system trend.
