为了确定局部浸润的肺单个核细胞(PMC)在清除下呼吸道病毒中的作用,作者研究了与氢氧化铝佐剂混合的纯化呼吸道合胞病毒(RSV)融合(F)蛋白(5μg)接种Balb/c小鼠的效果。 作者在0及3周对小鼠肌肉接种F蛋白以诱导保护性免疫力,同时与实验感染RSV(1~2×10~6 pfu)的小鼠进行比较,末次免疫或感染后3或4周用1×10~7pfu病毒作鼻内攻击,测定攻击后4和8天每克肺组织的病毒效价,并检测初次免疫后3和6周混合血清样本的中和抗体效价。 结果表明,F蛋白诱生的免疫应答与实验感染相似,能抑制攻击后4~8天的病毒复制,而未免疫对照小鼠在攻击后8天才有抑制作用。接种组和实验感染组于6周后查到抗RSV中和抗体。作者用~(51)Cr释放试验检测了初次接种后3周及RSV攻击后5和7天肺组织中浸润PMC的杀细胞作用,发现有抗原依
To determine the role of locally infiltrated lung mononuclear cells (PMCs) in clearance of lower respiratory tract virus, the authors studied the effect of Balb / c mice immunized with Purified Respiratory Syncytial Virus (RSV) fusion (F) protein (5 μg) / c mice. The authors intramuscularly inoculate F mice with Muscle F protein at 0 and 3 weeks to induce protective immunity, compared with mice experimentally infected with RSV (1-2 x 10-6 pfu), 3 or 4 weeks after last immunization or infection Intranasal challenge with 1 x 10 ~ 7 pfu virus was used to determine the virus titer per gram of lung tissue at 4 and 8 days post challenge as well as the neutralizing antibody titers of the pooled serum samples at 3 and 6 weeks after primary immunization. The results showed that the immune response induced by F protein was similar to the experimental infection and inhibited the virus replication 4 to 8 days after challenge, whereas the unimmunized control mice had inhibitory effect only 8 days after challenge. Anti-RSV neutralizing antibodies were found after 6 weeks in vaccinated and experimental infected groups.
