目的 观察离体条件下肿瘤患者外周血加热后T淋巴细胞亚群的改变。方法 将肝素化人外周血加至混合培养基中 ,分别放入不同温度水浴箱中孵育 3 0min ,然后在 3 7℃± 0 .5℃恒温条件下培养 72h。最后采用流式细胞仪 2 5 6道单参数直方图分析CD+ 4、CD+ 8、CD+ 56细胞值及CD+ 4/CD+ 8值。结果 鼻咽癌患者外周血在 40℃、42℃、43℃条件下加热 3 0min后 ,CD+ 4、CD+ 56细胞值显著上升 ,CD+ 8细胞值显著下降 ,CD+ 4/CD+ 8值上升 ;43℃条件下加热 3 0min后 ,与 42℃相比较 ,CD+ 4、CD+ 56细胞值有下降趋势 ,CD+ 8细胞值轻度上升 ,CD+ 4/CD+ 8值轻度下降。结论 外周血T淋巴细胞亚群不同的热敏感性可能是热疗调节机体免疫功能状态的机制之一。
Objective To observe the changes of T lymphocyte subsets in peripheral blood of tumor patients after heating in vitro. Methods Heparinized human peripheral blood was added to the mixed culture medium and incubated in a different temperature bath for 30 min, then cultured at 37°C±0.5°C for 72 h. Finally, flow cytometer 256 one-parameter histograms were used to analyze CD+4, CD+8, CD+56 cell values and CD+4/CD+8 values. Results After 30 min heating at 40°C, 42°C and 43°C in peripheral blood of patients with nasopharyngeal carcinoma, the values of CD+4 and CD+ 56 cells increased significantly, and the CD+ 8 cell value decreased significantly. The CD+ 4/CD+ 8 values increased; 43°C. After heating for 30 min under conditions, compared with 42°C, the values of CD+4 and CD+56 cells showed a decreasing trend, and the CD+8 cells increased slightly, while the CD+4/CD+8 values decreased slightly. Conclusion The different thermal sensitivity of T lymphocyte subsets in peripheral blood may be one of the mechanisms by which hyperthermia regulates the immune function of the body.
