目的探讨T-bet mRNA和GATA3 mRNA在正常妊娠产妇及子痫前期患者蜕膜组织中的表达差异。方法选取子痫前期和正常妊娠产妇作为实验组和对照组,采用RT-PCR方法,检测各组产妇蜕膜组织中T-bet mRNA和GA-TA3 mRNA表达水平。结果①子痫前期重度组与轻度组相比,子痫前期重度组与正常组相比T-bet mRNA表达量的差异有统计学意义(P<0.01,P<0.01)。子痫前期轻度组与正常组T-bet mRNA表达量相比较差异无统计学意义(P=0.089)。②子痫前期重度组与轻度组相比,子痫前期重度与正常组相比,GATA3 mRNA表达量的差异有统计学意义(P<0.01,P<0.01),子痫前期轻度组与正常组相比GATA3 mRNA表达量的差异无统计学意义(P=0.127)。③T-bet mRNA/GATA3 mRNA比值在正常组与子痫前期重度组间、子痫前期轻度组与重度组间差异有统计学意义(P<0.01,P<0.01)。二者比值在正常组与子痫前期轻度组间差异无统计学意义(P=0.522)。结论子痫前期Th1漂移可能与T-bet高表达和GATA3低表达有关。
Objective To investigate the expression differences of T-bet mRNA and GATA3 mRNA in normal pregnant women and preeclampsia patients. Methods The preeclampsia and normal pregnant women were selected as experimental group and control group. The expression of T-bet mRNA and GA-TA3 mRNA in decidua of each group were detected by RT-PCR. Results ① Compared with mild group, the expression of T-bet mRNA in severe preeclampsia group was significantly higher than that in mild group (P <0.01, P <0.01). The expression of T-bet mRNA in mild preeclampsia group and normal group had no significant difference (P = 0.089). Compared with the mild group, the level of GATA3 mRNA expression in severe preeclampsia group was significantly higher than that in mild group (P <0.01, P <0.01), and mild preeclampsia group There was no significant difference in GATA3 mRNA expression between normal group and normal group (P = 0.127). (3) The ratio of T-bet mRNA / GATA3 mRNA between normal group and severe preeclampsia group was significantly different between mild group and severe group (P <0.01, P <0.01). There was no significant difference between the two groups in the mild group and normal group (P = 0.522). Conclusion The pre-eclampsia Th1 drift may be related to the high expression of T-bet and the low expression of GATA3.
