观察铁离子螯合剂U74500A(U7A)和去铁胺(Desferrioxamine,Dfx)对内毒素血症小鼠急性肺损伤的保护作用。结果发现,U7A和Dfx能降低小鼠肺组织硫代巴比妥酸反应物(TBARS)含量及肺泡毛细血管通透性。体外实验发现,单纯铁离子能使正常大鼠肺、心浆TBARS产生增加,且呈明显量效关系;U7A对其抑制作用较Dfx强。提示U7A防治内毒素所致肺损伤可能与铁离子螯合,从而抑制铁依赖性脂质过氧化作用有关。
To observe the protective effect of iron chelator U74500A (U7A) and desferrioxamine (Dfx) on acute lung injury in endotoxemic mice. The results showed that U7A and Dfx can reduce the content of thiobarbituric acid reactant (TBARS) and the permeability of alveolar capillary in mouse lung tissue. In vitro experiments found that iron alone can make the normal rat lung, cardiac TBARS increased production, and showed a significant dose-effect relationship; U7A stronger inhibition than Dfx. These results suggest that the lung injury induced by U7A may be related to iron chelation and thus iron-dependent lipid peroxidation.
