目的研究磷脂酰肌醇3-激酶(PI3K)在卵巢癌组织中的表达及意义。方法(1)应用4 097靶基因点基因芯片对2006年吉林大学第一医院5例卵巢浆液性囊腺癌及5例正常卵巢组织进行基因表达谱分析,筛查卵巢癌差异表达基因。(2)应用RT-PCR检测2006年该院10例卵巢癌组织、10例卵巢瘤组织及10例正常卵巢组织中PI3K表达。结果(1)基因芯片筛查出卵巢浆液性囊腺癌差异表达基因58个,其中表达增高(上调趋势)基因30个,表达降低(下调趋势)基因28个,PI3K为表达上调基因。(2)PI3K基因在卵巢癌组织中表达增高,与正常卵巢及卵巢瘤比较差异有统计学意义(PI3KmRNA表达量分别为0.931±0.598、0.350±0.235、0.297±0.168,P<0.01),在卵巢瘤组织中表达与正常卵巢组织表达差异无统计学意义(P>0.05)。结论基因芯片可以用于筛查卵巢癌差异表达基因;PI3K为表达增高基因;PI3K在卵巢癌组织中表达显著高于卵巢瘤组织及正常卵巢组织。
Objective To study the expression and significance of phosphatidylinositol 3-kinase (PI3K) in ovarian cancer. Methods (1) Gene expression profiles of 5 ovarian serous cystadenocarcinomas and 5 normal ovarian tissues from the first hospital of Jilin University in 2006 were screened by 4 097 target gene microarray gene chips. The differentially expressed genes in ovarian cancer were screened. (2) The expression of PI3K in 10 cases of ovarian cancer tissues, 10 cases of ovarian tumor tissues and 10 cases of normal ovarian tissues in this hospital in 2006 was detected by RT-PCR. Results (1) 58 genes were differentially expressed in ovarian serous cystadenocarcinoma by gene chip. Among them, 30 were up-regulated (down-regulated), 28 were down-regulated (down-regulated) and PI3K was up-regulated. (2) The expression of PI3K gene in ovarian cancer tissues was significantly higher than that in normal ovarian tissues and ovarian tumors (PI3K mRNA levels were 0.931 ± 0.598,0.350 ± 0.235,0.297 ± 0.168, P <0.01) There was no significant difference in the expression between tumor tissue and normal ovarian tissue (P> 0.05). Conclusion Gene microarray can be used to screen differentially expressed genes in ovarian cancer. PI3K is a highly expressed gene. The expression of PI3K in ovarian cancer is significantly higher than that in ovarian cancers and normal ovarian tissues.
