目的 研究大鼠脑出血前脑内促凋亡基因半胱氨酰天冬氨酸特异性蛋白酶 - 3(caspase 3)的表达 ,以及三七总皂甙对其表达的影响。方法 用原位杂交法观察胶原酶诱导的脑出血大鼠前脑内caspase 3mRNA在给药组和对照组的表达变化 ;用免疫组化法观察caspase 3p2 0蛋白的变化。结果 脑出血后前脑内caspase 3mRNA及caspase 3p2 0主要在病灶及病灶周围、大脑皮质Ⅱ~Ⅴ层、海马CA2 、CA3区表达 ,阳性细胞包括变性神经元和胶质细胞。给药组阳性细胞数量显著减少 ,细胞着色变浅。结论 三七总皂甙可通过抑制caspase 3mRNA的转录及caspase 3蛋白的裂解活化 ,减少细胞凋亡 ,促进脑出血后前脑内神经元的存活及损伤修复
Objective To investigate the expression of caspase 3, a pro-apoptotic gene in the brain of rats with intracerebral hemorrhage, and the effect of panax notoginseng saponins on its expression. Methods The expression of caspase 3 mRNA in the forebrain of rats with collagenase-induced intracerebral hemorrhage was observed by in situ hybridization. The changes of caspase 3p2 0 protein were observed by immunohistochemistry. Results After intracerebral hemorrhage, caspase 3 mRNA and caspase 3p2 0 were mainly expressed in the lesions and around the lesion, in the cerebral cortex II-V layer, and in the hippocampal CA2 and CA3 regions. The positive cells included degenerated neurons and glial cells. The number of positive cells in the administration group was significantly reduced and the cells became lighter. Conclusion Panax notoginseng saponins can inhibit caspase 3 mRNA transcription and caspase 3 protein cleavage activation, reduce apoptosis, and promote the survival and injury repair of neurons in the forebrain after cerebral hemorrhage.
