研究壮骨颗粒含药血清对骨髓基质干细胞体外成骨活性的影响。方法:采用体外细胞培养技术,采用壮骨颗粒对细胞进行体外诱导培养,并设置空白血清对照组,进行MTT、免疫组织化学染色及ALP测定,观察壮骨颗粒对骨髓基质细胞的诱导作用。结果:在促细胞增殖方面:实验组作用显著(P<0.01),在第3d开始增殖,第6d达到平台期;而对照组对数增殖期明显延长(P<0.01);在促细胞分化方面:壮骨颗粒血清组在2d、3d、5dALP活性水平高于对照组(P<0.01)。常规形态学观察,壮骨颗粒组诱导的细胞在形态上类似于成骨细胞,免疫组化染色提示诱导细胞具有骨细胞表型。结论:壮骨颗粒组应用可促进细胞增殖,并诱导骨髓基质细胞向成骨细胞分化。
To study the effect of Zhuanggu granule-containing serum on osteogenic activity of bone marrow stromal stem cells in vitro. Methods: The cells were cultured in vitro using Zhuanggong granule by in vitro cell culture technique, and the blank serum control group was set up. MTT, immunohistochemical staining and ALP determination were performed to observe the effect of Zhuanggu granules on the induction of bone marrow stromal cells. Results: In the aspect of promoting cell proliferation, the effect of experimental group was significant (P <0.01), and began to proliferate on the third day and reach the plateau on the 6th day; while the logarithmic proliferation period of the control group was significantly prolonged (P <0.01) : The level of ALP activity of Zhuanggu granules serum group was higher than that of the control group on the 2nd, 3rd, 5th day (P <0.01). Conventional morphological observation showed that the cells induced by Zhuanggu granule group were similar in morphology to osteoblasts, and the immunohistochemical staining indicated that the induced cells had osteocyte phenotype. Conclusion: The application of Zhuanggu granules can promote cell proliferation and induce bone marrow stromal cells to differentiate into osteoblasts.
