采用聚丙稀酰胺凝胶电泳的方法, 对通过浸种法将外源DNA 直接导入所获得的烤烟变异体, 进行过氧化物酶同工酶谱分析。其结果是酶谱谱带清晰, 变异体与亲本的谱带有明显差异, 并与变异体的表型性状相一致。因此, 我们认为可以用同工酶分析法做为鉴定外源DNA导入烤烟的生化指标之一。应用浸种法将外源DNA导入烤烟, 可为烤烟良种繁育提供亲本和基础材料。
Using polyacrylamide gel electrophoresis, the foreign DNA was directly introduced into the obtained flue-cured tobacco by the soaking method, and the peroxidase isozymes were analyzed. The result is a clear zymogram band, a significant difference between the band and the parental band, and is consistent with the phenotypic traits of the mutant. Therefore, we think that isozyme assay can be used as one of the biochemical markers to identify the foreign DNA introduced into flue-cured tobacco. Introducing exogenous DNA into flue-cured tobacco by seed soaking method can provide parents and basic materials for flue-cured tobacco breeding.
