目的:探讨sigma-1(σ1)受体对情绪行为的调节作用。方法:采用旷场试验、悬尾试验、强迫游泳试验和Y迷宫的检测方法,分析5月龄雌和雄σ1受体基因敲除(σ1受体-KO)小鼠的抑郁样行为。用放射性免疫技术检测血浆的雌二醇和睾酮水平。结果:与野生型(WT)雄鼠相比,σ1受体-KO雄鼠在旷场中央区的运动时间明显减少,在悬尾试验和强迫游泳中的不动时间显著延长,在Y迷宫Novel臂的时间减少。但是,σ1受体-KO雌鼠的雌二醇水平和σ1受体-KO雄鼠的睾酮水平与对照组相比都没有改变。结论:σ1受体-KO诱发的抑郁样行为是雄鼠特异性的,可能与雌激素的抗抑郁作用有关。
Objective: To investigate the regulatory effect of sigma-1 (σ1) receptor on emotional behavior. Methods: Depression-like behaviors of 5-month-old male and female sigma 1 receptor knockout (σ1 receptor-KO) mice were analyzed using the open field test, the tail suspension test, the forced swimming test and the Y maze test. Radioimmunoassay was used to detect plasma estradiol and testosterone levels. Results: Compared with wild-type (WT) male rats, the sigma 1 receptor-KO male mice significantly reduced the movement time in the central open-field and prolonged immobility time in the tail-suspended test and forced swimming. Arm time reduced. However, estradiol levels in sigma 1 receptor -KO female mice and testosterone levels in sigma 1 receptor-ko male mice were unchanged from the control. Conclusion: The depression-like behaviors induced by sigma-1 receptor-KO are male-specific and may be related to the antidepressant effects of estrogen.
