目的:探索去甲肾上腺素对心肌细胞β受体影响的细胞内机制。方法:利用β-肾上腺素能受体放射性配基3H-DHA和Fura-2/AM测定心肌细胞受体和胞内游离钙。结果:(1)去甲肾上腺素可以引起心肌细胞β受体下调和胞内游离钙增高;(2)维拉帕米能抑制去甲肾上腺素诱导的β受体下调和胞内游离钙的增高。结论:去甲肾上腺素诱导心肌细胞内游离钙变化可能参与了心肌细胞β受体的调节
Objective: To explore the intracellular mechanism of norepinephrine on cardiomyocyte β receptor. Methods: 3H-DHA and Fura-2 / AM radioactive ligands of β-adrenergic receptor were used to determine cardiomyocyte receptor and intracellular free calcium. Results: (1) norepinephrine can cause downregulation of cardiomyocyte β receptor and increase of intracellular free calcium; (2) Verapamil can inhibit norepinephrine-induced β receptor downregulation and intracellular free calcium increase . CONCLUSION: Norepinephrine-induced changes in intracellular free calcium may be involved in the regulation of cardiomyocyte β receptors
