以油茶优良无性系“湘林1号”春季与夏季发生的新叶为外植体,在选定MS基本培养基的基础上,利用不同浓度的NAA与BA两种植物生长调节剂,对油茶叶片初始诱导培养阶段薄壁细胞的诱导与愈伤组织的发生进行了研究。结果表明,将春夏两季发生的新叶接种在MS+NAA(0.7 mg/L)+BA(2.0 mg/L)培养基中12~20 d后,得到了淡黄绿色的薄壁细胞与愈伤组织;52 d后,于黄白色的胚状体薄壁细胞中出现了再生的油茶茎叶。
Taking the new leaves of spring and summer in Camellia oleifera clone “Xianglin 1” as explants, on the basis of selected MS basal medium, different concentrations of NAA and BA plant growth regulators, The induction of parenchyma cells and the occurrence of callus in the initial induction of Camellia oleifera leaves were studied. The results showed that the yellowish green parenchyma cells were obtained after inoculated with MS + NAA (0.7 mg / L) and BA (2.0 mg / L) for 12-20 days in spring and summer. After 52 days, regenerated Camellia oleifera stems and leaves appeared in yellow-white embryoid body parenchyma cells.
