一、建立鼠杂交瘤12株,人杂交瘤2株。经体外连续培养一年以上,能稳定分泌鼠或人抗胃癌单抗,其中10株为IgG_1,1株IgG_2b、3株IgM。 二、在冰冻或石蜡切片染色,上述单抗不与正常胃粘膜等多种正常组织反应,联合染色在胃癌阳性率达95%以上。 三、相应抗原在细胞水平的分布可分为四型:即胞浆型、胞膜型、胞外型及混合型。 四、相应抗原在亚细胞水平的定位主要在胞膜或其特化结构微绒毛上,或胞浆内的内质
First, 12 mouse hybridomas and 2 human hybridomas were established. After being continuously cultured in vitro for more than one year, it can stably secrete murine or human anti-gastric monoclonal antibodies, of which 10 are IgG_1, 1 IgG_2b, and 3 IgM. Second, in freezing or paraffin section staining, the above-mentioned monoclonal antibody does not react with normal tissues such as normal gastric mucosa, and the positive rate of combined staining in gastric cancer is more than 95%. Third, the distribution of the corresponding antigens at the cellular level can be divided into four types: cytoplasm, cell membrane type, extracellular type, and mixed type. 4. The localization of the corresponding antigen at the subcellular level is mainly on the membrane or its specialized structure, microvilli, or intracytoplasmic endoplasm.
