目的:探讨血根碱对S180荷瘤小鼠的抑瘤作用及对血管生成的影响。方法:建立小鼠S180皮下移植瘤模型,随机分为模型组、环磷酰胺(CTX 20 mg/kg)组、血根碱(10、20、40 mg/kg)组、联合组(血根碱10 mg/kg+CTX20 mg/kg),连续给药10 d后处死小鼠,检测移植瘤质量及体积并计算抑瘤率;HE染色观察肿瘤细胞的形态学特征;免疫组化法检测药物对微血管密度(MVD)、血管内皮生长因子(VEGF)表达的影响。结果:与模型组比较,CTX组、血根碱(20、40 mg/kg)组、联合用药组肿瘤生长显著受到抑制;肿瘤组织HE染色显示,与模型组比较给药组中肿瘤细胞异型性降低,病理性核分裂减少,可见散在及融合成片状的坏死灶;免疫组化显示CTX、血根碱(20、40mg/kg)及联合用药均能显著减少MVD、VEGF的表达,联合用药时抑瘤效果更显著(P<0.01,P<0.05)。结论:血根碱可通过下调S180移植瘤组织中MVD、VEGF的表达,从而抑制肿瘤血管的生成发挥抗肿瘤作用。
Objective: To investigate the anti-tumor effect of sanguardine on S180 tumor-bearing mice and its effect on angiogenesis. Methods: A subcutaneous xenograft model of S180 mice was established and randomly divided into model group, cyclophosphamide (CTX 20 mg / kg) group, sanguinarine (10,20,40 mg / kg) group, combination group 10 mg / kg + CTX20 mg / kg). The mice were sacrificed 10 days after continuous administration, the quality and volume of the transplanted tumor were detected and the tumor inhibition rate was calculated. Results: Compared with model group, tumor growth in CTX group, sanguinarine (20,40 mg / kg) group and combined treatment group was significantly inhibited. HE staining of tumor tissue showed that compared with model group, tumor cell atypia Decreased, pathological mitosis decreased, scattered and fused into pieces of necrotic lesions can be seen; immunohistochemistry CTX, sanguinarine (20,40 mg / kg) and the combination of drugs can significantly reduce the expression of MVD, VEGF, when combined Anti-tumor effect was more significant (P <0.01, P <0.05). Conclusion: Scumenditetine can inhibit tumor angiogenesis by down-regulating the expression of MVD and VEGF in S180 xenografts.
