目的:研究地黄饮子的益智作用。方法:应用大鼠跳台实验、小鼠水迷宫实验观察记忆能力,原位杂交法检测神经细胞NMDA-2AmRNA及NMDAR1mRNA表达。结果:给予地黄饮子的MSG动物与模型组比较,跳台错误反应次数显著减少,触电潜伏期明显延长,平台撤除后,有搜寻平台的目的性,CPP较模型组显著升高(P<0.01),脑组织中Glu含量显著升高,(P<0.01),Glu/GABA值有增高的趋势;海马NMDAR1、NR2 mRNA免疫反应物光密度较MSG组显著升高(P<0.01)。结论:地黄饮子能增强MSG大鼠神经原NMDAR1和NMDAR2表达,改善MSG大鼠学习记忆功能。
Objective: To study the puzzle effect of Dihuang Yinzi. METHODS: Rats’ step-down test and mouse water maze test were used to observe the memory ability. In situ hybridization was used to detect the expression of NMDA-2A mRNA and NMDAR1 mRNA in nerve cells. RESULTS: Compared with the model group, MSG animals given Dihuangyinzi significantly decreased the number of false reaction times on the platform and the latency of electrocution was significantly prolonged. After the platform was removed, there was a purpose of the search platform, and CPP was significantly higher than that of the model group (P<0.01). The Glu content in brain tissue increased significantly (P<0.01), and the Glu/GABA value increased; the optical density of NMDAR1 and NR2 mRNA in the hippocampus was significantly higher than that of MSG group (P<0.01). Conclusion: Dihuang Yinzi can increase the expression of neuronal NMDAR1 and NMDAR2 in MSG rats and improve the learning and memory function of MSG rats.
