目的:观察王氏连朴饮对脾胃湿热证大鼠血清白细胞介素1(IL-1β)和一氧化氮(NO)含量的调节作用。方法:将30只大鼠随机分为正常对照组、脾胃湿热证模型组、治疗组三组,每组10只。除正常对照组外,其余各组均按“湿热环境+高脂高糖饮食+白酒”的内外因湿热证模型综合造模方法,建立脾胃湿热证大鼠模型。放免法检测IL-1β含量,硝酸还原酶法检测NO含量。结果:脾胃湿热证模型大鼠血清中IL-1β含量明显增高,NO含量显著降低(与正常对照组相比有显著性差异P<0.01),经治疗后大鼠血清中IL-1β含量、NO含量逐渐恢复正常,与正常对照组相比无显著性差异(P<0.01)。结论:王氏连朴饮能有效改善大鼠脾胃湿热证诸症状,使增高的血清IL-1β含量恢复正常,使降低的血清NO含量恢复正常。提示其作用机制可能是:通过调节脾胃湿热证大鼠血清中NO的含量,恢复抗氧化功能,调节大鼠的免疫功能;通过调节脾胃湿热证大鼠血清中IL-1β的含量,减少致热源,减轻炎症反应,从而取得治疗脾胃湿热证的疗效。
Objective: To observe the effect of Wangshi Lian Puyin on the levels of serum interleukin-1 (IL-1β) and nitric oxide (NO) in rats with spleen-stomach dampness-heat syndrome. Methods: Thirty rats were randomly divided into normal control group, spleen-stomach damp-heat syndrome group, and treatment group. There were 10 rats in each group. In addition to the normal control group, the rest of the groups were established according to the “humidity and heat environment + high-fat high-glucose diet + liquor ” model of internal and external heat-damp heat syndrome model to establish a rat model of spleen-stomach dampness-heat syndrome. Radioimmunoassay was used to detect IL-1β content, and nitrate reductase assay was used to detect NO content. Results: The levels of IL-1β in serum of rats with spleen-stomach dampness-heat syndrome were significantly higher, and the content of NO was significantly decreased (significant difference compared with normal control group P<0.01). IL-1β content in serum of rats after treatment was negative. The content gradually returned to normal, with no significant difference compared with the normal control group (P <0.01). Conclusion: Wangshi Lian Puyin can effectively improve various symptoms of spleen-stomach damp-heat syndrome in rats, increase the serum IL-1β content to normal, and make the reduced serum NO content return to normal. It suggests that the mechanism may be: by regulating the NO content in the serum of rats with spleen-stomach dampness-heat syndrome, restoring anti-oxidation function, regulating the immune function of rats; by regulating the serum IL-1β content in rats with spleen-stomach dampness-heat syndrome, reducing the heat source , To reduce the inflammatory response, so as to achieve therapeutic effect of spleen and stomach dampness and heat syndrome.
