：
目的 通过网络药理学的方法初步探讨温肺疏风方治疗咳嗽变异性哮喘的作用机制.方法 基于中药系统药理学数据库与分析平台(TCMSP)数据库筛选温肺疏风方中各味中药的主要化学成分及相应靶点,通过蛋白质数据库筛选出物种为“人”的靶点.从Drugbank、OMIM、KEGG及DisGeNET数据库中检索哮喘相关靶点,将药物靶点与哮喘相关靶点进行映射得到温肺疏风方作用于哮喘的预测靶点.采用String数据库构建温肺疏风方作用于哮喘的体内反应网络图并筛选出重要作用关系,利用Cytoscape软件构建蛋白相互作用(PPI

目的 通过网络药理学的方法初步探讨温肺疏风方治疗咳嗽变异性哮喘的作用机制.方法 基于中药系统药理学数据库与分析平台(TCMSP)数据库筛选温肺疏风方中各味中药的主要化学成分及相应靶点,通过蛋白质数据库筛选出物种为“人”的靶点.从Drugbank、OMIM、KEGG及DisGeNET数据库中检索哮喘相关靶点,将药物靶点与哮喘相关靶点进行映射得到温肺疏风方作用于哮喘的预测靶点.采用String数据库构建温肺疏风方作用于哮喘的体内反应网络图并筛选出重要作用关系,利用Cytoscape软件构建蛋白相互作用(PPI)网络,进行拓扑分析得到关键靶点.通过DAVID数据库进行KEGG信号通路富集分析.结果 共获得97个化合物,239个靶点,哮喘相关靶点共1 413个.疾病靶点与药物靶点映射得到110个温肺疏风方作用于哮喘的靶点,PPI网络拓扑分析得到JUN、STAT3、QBHIJ4、TNF、EDN1、MAPK等17个关键蛋白,通路富集分析得到包括TNF信号通路、Toll样受体信号通路、MAPK信号通路、c AMP信号通路、NOD样受体信号通路等11条富集程度较高的通路.结论 温肺疏风方可能主要通过抗炎发挥治疗哮喘的作用,TLR4-NFκB/MAPK通路是其发挥作用的关键通路之一.
