研究碱化法结合大孔树脂法纯化贯叶连翘总黄酮的工艺条件。方法:以静态饱和吸附量、静态解析率为指标对6种大孔树脂进行了筛选;又以总黄酮含量为指标对提取液上柱前的预处理方式、树脂吸附工艺参数进行了优选。结果:D101大孔树脂吸附量、解析率最好;上样液浓度为0.2 g生药/mL;上样速度为2BV/h;洗脱剂为60%乙醇;洗脱速度为3BV/h;洗脱剂用量为5BV/h。结论:优选的贯叶金丝桃总黄酮纯化工艺稳定、简便可行、适合大生产。
Study on the alkaline conditions combined with macroporous resin purification of the total flavonoids of Hypericum perforatum conditions. Methods: Six kinds of macroporous resins were screened based on static adsorption capacity and static analysis rate. The optimum pretreatment method and resin adsorption process parameters of the extract were also optimized with total flavonoids content. Results: D101 macroporous resin adsorption capacity, the best resolution; the sample concentration of 0.2 g crude drug / mL; loading speed 2BV / h; eluent 60% ethanol; elution speed 3BV / h; wash The amount of removing agent is 5BV / h. Conclusion: The optimized process of purification of total flavonoids from Hypericum perforatum is stable, simple and feasible and suitable for large-scale production.
