目的:建立测定桔梗冬花片中桔梗皂苷D的HPLC-ELSD方法。方法:色谱柱:Diamonsil C_(18)(200 mm×4.6 mm,5μm),流动相:乙腈-水(26:74),流速:1.0 ml·min~(-1);Waters 2424蒸发光散射检测器检测,漂移管温度85℃,气体流速为2.9SLPM·min~(-1)。结果:桔梗皂苷D的进样量在1.010~12.625μg范围内,线性关系良好(r=0.999 4),平均回收率为97.92%,RSD=2.05%(n=6)。结论:本法可用于测定桔梗冬花片中桔梗皂苷D含量,方法准确、可靠,操作简便。
OBJECTIVE: To establish a HPLC-ELSD method for the determination of platycodin D in Ophiopogon japonicus. METHODS: Diamonsil C_ (18) (200 mm × 4.6 mm, 5 μm) was used as the mobile phase. The mobile phase consisted of acetonitrile and water (26:74) and the flow rate was 1.0 ml · min -1. The temperature of the drift tube was 85 ℃ and the gas flow rate was 2.9 SLPM · min ~ (-1). Results: The calibration curve of Platycodin S was in the range of 1.010 ~ 12.625 μg with a good linearity (r = 0.999 4). The average recovery was 97.92% and the RSD was 2.05% (n = 6). Conclusion: This method can be used to determine the content of platycodin D in Ophiopogon japonicus. The method is accurate, reliable and easy to operate.
