目的 :研究活化人外周血单个核细胞(peripheral blood mononuclear cell,PBMC)在体外影响口腔鳞癌细胞生长及凋亡的机制。方法:用酶联免疫吸附试验(ELISA)、反转录聚合酶链反应(RT-PCR)、四甲基偶氮唑蓝(MTT)法以及流式细胞仪(FCM)检测植物血凝素(phytohemagglutinin,PHA)刺激前、后人PBMC分泌细胞因子变化及其对口腔鳞癌细胞生长与凋亡的影响。采用SPSS17.0软件包对数据进行统计学分析。结果:以PHA刺激人PBMC为实验组分泌的细胞因子显著高于对照组(P<0.01);实验组能够诱导颗粒酶B及穿孔素m RNA的表达;实验组抑制口腔肿瘤细胞生长及诱导口腔鳞癌细胞凋亡也显著增加(P<0.05)。结论:PHA激活的人PBMC在体外能够显著抑制口腔鳞癌细胞生长及诱导口腔鳞癌细胞凋亡。
Objective: To investigate the mechanism of activating peripheral blood mononuclear cells (PBMCs) on the growth and apoptosis of oral squamous cell carcinoma in vitro. Methods: The contents of phytohemagglutinin (IL-6), leptin and their receptor were detected by enzyme linked immunosorbent assay (ELISA), reverse transcription polymerase chain reaction (RT-PCR), MTT assay and flow cytometry (FCM) Effects of Phytohemagglutinin and PHA on the Secretion of Cytokines in PBMCs and Their Effects on the Growth and Apoptosis of Oral Squamous Cell. SPSS17.0 software package for statistical analysis of the data. Results: The cytokines secreted by PHA stimulated human PBMC were significantly higher than that of control group (P <0.01). The experimental group was able to induce the expression of granzyme B and perforin m RNA. The experimental group inhibited the growth of oral tumor cells and induced the oral cavity The apoptosis of squamous cell carcinoma was also significantly increased (P <0.05). CONCLUSION: PHA-activated human PBMC can significantly inhibit the growth of oral squamous cell carcinoma and induce the apoptosis of oral squamous cell carcinoma in vitro.
