目的:应用microRNAs芯片筛选肾透明细胞癌细胞差异表达microRNAs,并寻找差异表达microRNAs调控的靶基因,为进一步研究其在肾透明细胞癌发病机制中的作用打下基础。方法:应用哺乳动物microRNAs表达谱芯片检测11位肾透明细胞癌患者癌组织及非肿瘤肾组织中microRNAs差异表达。结果:81种microRNAs在肾透明细胞癌组中有效表达,其中48种microRNAs仅在肾透明细胞癌组中有效表达;42种microRNAs在对照组中有效表达,9种microRNAs仅在对照组中有效表达。33种microRNAs在两组中均表达,其中19种microRNAs表达分析有显著差异。芯片显示结果经northern blot分析确认。结论:microRNAs在肾透明细胞癌细胞中异常表达,提示它们可能在肾透明细胞癌的发生发展中起着重要的调控作用。
OBJECTIVE: To screen differentially expressed microRNAs in renal clear cell carcinoma cells by microRNAs and to find target genes regulated by differentially expressed microRNAs, so as to lay the foundation for further study on its role in the pathogenesis of clear cell renal cell carcinoma. Methods: Mammalian microRNAs microarray was used to detect differential expression of microRNAs in cancerous and non-cancerous renal tissues of 11 patients with clear cell renal cell carcinoma. Results: 81 kinds of microRNAs were effectively expressed in renal clear cell carcinoma group, of which 48 kinds of microRNAs were only effectively expressed in renal clear cell carcinoma group; 42 kinds of microRNAs were effectively expressed in control group and 9 kinds of microRNAs were only expressed in control group . 33 microRNAs were expressed in both groups, of which 19 microRNAs expression analysis were significantly different. Chips show the results confirmed by northern blot analysis. Conclusion: The abnormal expression of microRNAs in renal clear cell carcinoma cells suggests that they may play an important regulatory role in the development of renal clear cell carcinoma.
