目的 研究局灶性脑缺血再灌注期 ,一氧化氮合酶 (NOS)与髓过氧化物酶 (MPO)之间的关系。方法 采用大鼠大脑中动脉线栓 /再灌注模型 ,根据不同再灌注时间对大鼠分组 ,同时设定假手术组 ,取缺血区相邻脑组织分别检测NOS和MPO活性 ,对二者进行相关性分析。结果 假手术组可测得一定的NOS活性 ,再灌注早期 ( 1h)在缺血相邻区即可检测到一定的NOS活性 ,以后随着再灌注时间延长 ,测得NOS活性逐渐升高 ,再灌注 2 4h活性达最高。而假手术组及再灌注早期 ( 1h)代表白细胞浸润程度的MPO活性几乎测不到 ,再灌注早期 6h开始可测到一定的MPO活性 ,以后随着再灌注时间延长 ,测得MPO活性逐渐升高 ,再灌注 2 4hMPO活性达最高。NOS和MPO呈高度正相关。结论 在局灶性脑缺血 /再灌注期NO与白细胞浸润具有相关性
Objective To study the relationship between nitric oxide synthase (NOS) and myeloperoxidase (MPO) during focal cerebral ischemia / reperfusion. Methods The model of middle cerebral artery occlusion and reperfusion in rats was established. The rats were divided into groups according to the different reperfusion time. At the same time, the sham-operation group was set up. The activities of NOS and MPO were detected in adjacent brain tissue in ischemic area, Correlation analysis. Results The NOS activity was measured in the sham operation group. Some NOS activity was detected in the ischemic area in the early reperfusion period (1h), and then the activity of NOS was gradually increased with the prolongation of reperfusion time 24 h perfusion reached the highest activity. In the sham operation group and the early reperfusion (1h), the MPO activity that represented the degree of leukocyte infiltration was almost undetectable. Some MPO activity was detected at 6h after reperfusion, and then the MPO activity was gradually increased with the reperfusion time High, reperfusion 24 hMPO activity reached the highest. NOS and MPO were highly correlated. Conclusion There is a correlation between NO and leukocyte infiltration during focal cerebral ischemia / reperfusion
