目的建立HPLC法测定止咳合剂中吗啡的含量。方法色谱柱:Wonda Sil C18(4.6 mm×150 mm,5μm);流动相:0.01 mol·L-1磷酸二氢钾溶液-0.0025 mol·L-1庚烷磺酸钠溶液(含0.1%三乙胺,磷酸调p H至2.5)-乙腈(90∶10);流速:1.0 ml·min-1;检测波长:220 nm;柱温:30℃;进样量10μl。结果吗啡在0.525~42.0μg·ml-1范围内线性关系良好(r=1.0000),平均回收率为99.97%,RSD为1.27%(n=9)。结论本方法简便准确,重复性好,能够用于止咳合剂中吗啡的含量测定和质量控制。
Objective To establish an HPLC method for the determination of morphine in Zhike Mixture. Methods The mobile phase was Wonda Sil C18 (4.6 mm × 150 mm, 5 μm). The mobile phase consisted of 0.01 mol·L-1 potassium dihydrogen phosphate solution and 0.0025 mol·L -1 sodium heptane sulfonate solution containing 0.1% Amine, adjusted to pH 2.5 with phosphoric acid) -acetonitrile (90:10); flow rate: 1.0 ml · min-1; detection wavelength: 220 nm; column temperature: 30 ° C; Results Morphine showed a good linear relationship (r = 1.0000) in the range of 0.525 ~ 42.0μg · ml-1 with an average recovery of 99.97% and a RSD of 1.27% (n = 9). Conclusion The method is simple, accurate and reproducible. It can be used for the determination and quality control of morphine in Cough-Reliever Mixture.
