目的:探讨自杀性肿瘤疫苗(FC/TK)对大鼠卵巢癌的治疗作用。方法:采用聚乙二醇法,将自然杀伤基因修饰的卵巢癌细胞与树突状细胞融合制备成FC/TK,选取40只雌性Fischer344大鼠,制备Fischer344大鼠卵巢癌荷瘤模型,采用随机数字表法分为FC/TK组(FC/TK处理)、未经自然杀伤基因修饰的卵巢癌细胞与树突状细胞融合的疫苗(FC)处理组(FC组)、经60Co照射的FC/TK处理组(60Co-FC/TK组)和对照组(等量PBS处理)各10只。计算各组大鼠90天后的肿瘤平均重量、抑瘤率等指标。结果:FC组、FC/TK组的瘤体重量(3.17±0.68)g、(2.92±0.59)g均显著低于60Co-FC/TK组和PBS组且差异均具有统计学意义(P<0.05);FC组、FC/TK组的抑瘤率(61.53±10.28)%、(64.56±11.77)%显著高于60Co-FC/TK组且差异具有统计学意义(P<0.05)。FC组、FC/TK组的血清IL-2、TNF-ɑ水平显著低于60Co-FC/TK组和PBS组且差异均具有统计学意义(P<0.05)。FC组、FC/TK组的瘤体实质中CD4+、CD4+/CD8+比例显著的高于60Co-FC/TK组和PBS组,CD8+细胞显著的低于60Co-FC/TK组和PBS组且差异均具有统计学意义(P<0.05)。结论:FC/TK对大鼠卵巢癌具有显著的治疗作用,可能和提高肿瘤实质内部的T淋巴细胞亚群水平、降低血清炎症因子水平有关。
Objective: To investigate the therapeutic effect of suicide cancer vaccine (FC / TK) on ovarian cancer in rats. Methods: 40 female Fischer344 rats were prepared by fusion of natural killer gene-modified ovarian cancer cells and dendritic cells using polyethylene glycol method. FCM was used to prepare Fischer344 ovarian cancer tumor model. The digital meter method was divided into the FC / TK group (FC / TK group), the FC group (FC group) which was not fused with dendritic cells modified by natural killer gene, FC / TK group (60Co-FC / TK group) and control group (equal amount of PBS group). Calculate the average weight of tumor in each group of rats after 90 days, inhibition rate and other indicators. Results: The tumor weight (3.17 ± 0.68) g and (2.92 ± 0.59) g in FC group and FC / TK group were significantly lower than those in 60Co-FC / TK group and PBS group ). The tumor inhibition rates in FC group and FC / TK group were 61.53 ± 10.28% and 64.56 ± 11.77%, respectively, which were significantly higher than those in 60Co-FC / TK group (P <0.05). The serum levels of IL-2 and TNF-ɑ in FC / TK group were significantly lower than those in 60Co-FC / TK group and PBS group (P <0.05). The percentages of CD4 + and CD4 + / CD8 + in tumor cells in FC group and FC / TK group were significantly higher than those in 60Co-FC / TK group and PBS group. CD8 + cells were significantly lower than those in 60Co-FC / TK group and PBS group Statistically significant (P <0.05). Conclusion: FC / TK has a significant therapeutic effect on ovarian cancer in rats and may be related to increasing the level of T lymphocyte subsets and decreasing the level of serum inflammatory cytokines in tumor parenchyma.
