背景:电磁辐射在应用到许多领域的同时,对人体健康也构成一定的威胁。男性生殖系统也是电磁辐射的敏感靶器官,睾酮在电磁辐射条件下的变化及其机制目前还不清楚。目的:探讨电磁辐射对成年大鼠睾丸组织中类固醇合成急性调节蛋白(StAR)、细胞色素P450胆固醇侧链裂解酶(P450scc)的作用,初步揭示电磁辐射影响睾酮合成的分子机制。设计:随机对照研究。单位:解放军第三军医大学大坪医院野战外科研究所核医学科和劳动卫生学教研室。材料:本研究于2003-06/2004-05在解放军第三军医大学大坪医院野战外科研究所核医学科和劳动卫生学教研室完成。选择二级清洁级雄性成年Wistar大鼠75只,随机分为5组,即对照组以及电磁辐射后3,6,24,72h组,每组15只,体质量180~220g。方法:采用峰值功率为90W/cm2的微波连续辐照15min,辐照完毕后,动物均存活。采用放射免疫方法(RIA)分别测定电磁辐照后3,6,24,72h及对照组血清睾酮含量,同时运用RT-PCR方法测定睾丸组织中StAR、P450sccmRNA水平。主要观察指标:电磁辐照后3,6,24,72h组与对照组血清睾酮含量和StAR,P450sccmRNA的水平。结果:辐照后3h血清睾酮含量和StAR、P450sccmRNA水平均明显低于对照组,分别较对照组降低83.1%,57.3%,53.6%(P<0.01);6h略有回升,但仍明显低于对照组,分别较?
Background: Electromagnetic radiation, while applied to many fields, also poses a threat to human health. The male reproductive system is also a sensitive target organ of electromagnetic radiation. The change of testosterone under electromagnetic radiation and its mechanism are not yet clear. Objective: To investigate the effect of electromagnetic radiation on steroidogenic acute regulatory protein (StAR), cytochrome P450 cholesterol side chain lyase (P450scc) in adult rat testis and to reveal the molecular mechanism of electromagnetic radiation on testosterone synthesis. Design: Randomized controlled study. Unit: Institute of Field Surgery, Daping Hospital, Third Military Medical University. Department of Nuclear Medicine and Department of Labor Hygiene. MATERIALS: The study was performed at the Department of Nuclear Medicine and Labor Hygiene, Institute of Field Surgery, Daping Hospital, Third Military Medical University from June 2003 to May 2004. A total of 75 adult male Wistar rats of secondary clean grade were randomly divided into 5 groups: control group and 3, 6, 24 and 72 h after EMF. Methods: The microwaves with the peak power of 90W / cm2 were irradiated continuously for 15min. After irradiation, the animals survived. Radioimmunoassay (RIA) was used to measure the levels of serum testosterone at 3, 6, 24 and 72 hours after the electromagnetic irradiation and the control group respectively. Meanwhile, the levels of StAR and P450sccmRNA in testis were determined by RT-PCR. MAIN OUTCOME MEASURES: Serum testosterone levels and levels of StAR and P450sccmRNA at 3, 6, 24 and 72 h after irradiation with electromagnetic radiation.
