采用改良的分光光度法及豌豆凝集素亲和免疫电泳同步测定90例肝细胞癌(HCC)血清α-L-岩藻糖苷酶(AFU)活性和岩藻糖基化甲胎蛋白(AFP)含量。相关分析表明,HCC血清AFU活性与岩藻糖基化AFP相对含量无关(r=0.099),与岩藻糖基化AFP绝对含量亦无关(r=0.23)。将90例HCC按岩藻糖基化AFP相对含量不同(以25%为界)分组,两组血清AFU活性分别为535±220 nmol·ml~(-1)·h~(-1)和550±180 nmol·ml~(-1)·h~(-1),相差不显著(P>0.05)。若以血清AFU活性高低(以550 nmol·ml~(-1)·h~(-1)为界)分组,两组血清岩藻糖基化AFP相对含量分别为40%±23%和45%±22%,差异亦不显著(P>0.05)。提示,肝脏病变时AFP糖链结构的变化不是血清AFU参与的结果,可能来自于糖链生物合成途径的改变。
Simultaneous Determination of α-L-Fucosidase (AFU) Activity and Fucosylated Alpha-fetoprotein (AFP) Content in 90 Cases of Hepatocellular Carcinoma (HCC) Using a Modified Spectrophotometric Method and Pealectin Affinity Immunoelectrophoresis . Correlation analysis showed that the serum AFU activity of HCC was not related to the relative content of fucosylated AFP (r=0.099), nor was it related to the absolute content of fucosylated AFP (r=0.23). In 90 cases of HCC, the relative content of fucosylated AFP was different (with 25% as the boundary). The serum AFU activity in the two groups was 535±220 nmol·ml -1 ·h -1 and 550 respectively. ±180 nmol·ml~(-1)·h~(-1), with no significant difference (P>0.05). If serum AFU activity is divided into groups (with 550 nmol·ml -1 ·h -1), the relative content of serum fucosylated AFPs in the two groups is 40%±23% and 45%, respectively. ±22%, the difference was not significant (P>0.05). It is suggested that the changes in the AFP sugar chain structure during hepatic lesions are not the result of serum AFU involvement and may be due to changes in the sugar chain biosynthetic pathway.
