目的观察伤寒Vi多糖结合疫苗生产用菌株伤寒沙门菌CMCC50098株和副伤寒甲沙门菌CMCC50073株毒性和抗原性的稳定性。方法将CMCC50098和CMCC50073菌株分别连续传代至30代,取3、5、10、15、20、25、30代次菌,进行小鼠毒性试验,免疫家兔制备血清,进行抗原性试验。结果CMCC50098和CMCC50073菌株连续传30代,毒性均未改变,抗原性均未下降,血清凝集效价均达到1∶12800。结论CMCC50098和CMCC50073菌株连续传代至30代,毒性及抗原性均稳定。
Objective To observe the toxicity and antigenicity of strains Salmonella typhi CMCC50098 and Salmonella paratyphi CMCC50073 for the production of typhoid Vi polysaccharide conjugate vaccine. Methods The strains CMCC50098 and CMCC50073 were respectively passaged serially to 30 generations. The mice were sacrificed at 3, 5, 10, 15, 20, 25 and 30 times for toxicity test. The rabbits were immunized to prepare serum for antigenicity test. Results The strains CMCC50098 and CMCC50073 were serially passaged for 30 passages, with no change in toxicity and no decrease in antigenicity. The serum agglutination titer reached 1: 12800. Conclusion The strains CMCC50098 and CMCC50073 were passaged continuously to 30 generations with stable toxicity and antigenicity.
