目的:制备苏丹红Ⅰ完全抗原。方法:根据苏丹红Ⅰ(Sudan-Ⅰ)分子特点,合成Sudan-Ⅰ结构类似物,与Su-dan-Ⅰ结构相比,该结构类似物在其偶氮环对位引进一个羧基。通过碳二亚胺法(EDC)与牛血清蛋白(BSA)载体蛋白偶联,偶联产物经透析纯化后,用SDS凝胶电泳、紫外扫描鉴定,Sudan-Ⅰ结构类似物与BSA的平均偶联比达到2~3个。用该偶联物免疫小鼠,获得抗苏丹红Ⅰ血清。结果:成功制备了苏丹红Ⅰ完全抗原。结论:通过合成结构类似物,与大分子物质偶联,获取抗苏丹红Ⅰ血清是可行的。
Objective: To prepare Sudan Ⅰ complete antigen. Methods: According to the molecular characteristics of Sudan Ⅰ, the structural analogues of Sudan-Ⅰ were synthesized. Compared with the structure of Su-dan-Ⅰ, this structural analogue introduced a carboxyl group in the para-azo ring. Coupled with bovine serum albumin (BSA) carrier protein by carbodiimide method (EDC), the coupled product was purified by dialysis and identified by SDS gel electrophoresis and UV-scan. The average even number of Sudan-I analogues and BSA United ratio of 2 to 3. Mice were immunized with this conjugate to obtain anti-Sudan I serum. Results: Sudan Ⅰ complete antigen was successfully prepared. Conclusion: It is feasible to synthesize structural analogs and conjugate with macromolecules to obtain anti-Sudan Ⅰ serum.
