目的:建立HPLC测定维A酸在小鼠骨髓中含量的方法。方法:色谱柱为Diamonsil C18柱(150mm×4.6 mm,5μm),流动相为甲醇-水-冰醋酸(95∶5∶0.5),流速为1.0 mL.min-1,检测波长为350 nm。外标法定量。结果:维A酸的线性范围为0.01~10μg.mL-1(r=0.999 9),最低检测限为0.5 ng。日间与日内精密度均<5%。低、中、高3个浓度的平均绝对回收率为74.5%,方法回收率为99.3%。结论:该方法灵敏度高、准确,可用于小鼠骨髓组织中维A酸浓度的测定。
Objective: To establish a HPLC method for the determination of retinoic acid in bone marrow of mice. METHODS: Diamonsil C18 column (150 mm × 4.6 mm, 5 μm) was used as the mobile phase. The mobile phase was methanol-water-glacial acetic acid (95: 5: 0.5) at a flow rate of 1.0 mL · min- External standard method. RESULTS: The linear range of retinoids was 0.01-10 μg.mL-1 (r = 0.999 9) with the lowest detection limit of 0.5 ng. Day and day precision were <5%. The average absolute recovery of low, medium and high concentrations of three concentrations was 74.5%, the method recovery was 99.3%. Conclusion: The method is sensitive and accurate and can be used for the determination of retinoids in mouse bone marrow.
