目的建立高效液相色谱法测定葛根解酒片中葛根素和2,3,5,4’-四羟基二苯乙烯-2-O-β-D-葡萄糖苷(以下简称二苯乙烯苷)的含量。方法采用kromasil C18柱(250 mm×4.6 mm,5μm),流动相:乙腈(A)-水(B),梯度洗脱〔0 min,φ(B)=90%、40 min,φ(B)=85%、60 min,φ(B)=75%〕,流速:1.0 mL.min-1,检测波长:250 nm(0 min)、320 nm(30 min)。结果葛根素和二苯乙烯苷进样量分别为50.7~676.0 mg.L-1(r=0.999 6)和13.8~184.0 mg.L-1(r=0.999 5)线性关系良好,平均回收率分别为96.7%(RSD=1.7%,n=6)和96.4%(RSD=1.4%,n=6)。结论该方法可同时测定葛根解酒片中葛根素和二苯乙烯苷的含量。
OBJECTIVE To establish a HPLC method for the determination of puerarin and 2,3,5,4’-tetrahydroxystilbene-2-O-β-D-glucoside (hereinafter referred to as stilbene glycoside) content. Methods The mobile phase consisted of acetonitrile (A) - water (B), gradient elution [0 min, φ (B) = 90%, 40 min and φ (B)) on a Kromasil C18 column (250 mm × 4.6 mm, = 85%, 60 min, φ (B) = 75%]. The flow rate was 1.0 mL.min-1. The detection wavelength was 250 nm (0 min) and 320 nm (30 min). Results The calibration curves of puerarin and stilbene glycosides were linear in the range of 50.7-676.0 mg.L-1 (r = 0.999 6) and 13.8-184.0 mg.L-1 (r = 0.999 5), respectively. The mean recoveries were 96.7% (RSD = 1.7%, n = 6) and 96.4% (RSD = 1.4%, n = 6). Conclusion This method can be used to determine puerarin and stilbene glycosides in Puerarin.
