目的探讨Survivin在柔红霉素(DNR)与长春新碱(VCR)作用机制中的作用。方法2005年在武汉同济医院儿科血液实验室,选用高表达Survivin的Molt-4细胞,流式细胞术检测DNR、VCR作用后Molt-4细胞的凋亡,逆转录-聚合酶链反应(RT-PCR)检测DNR、VCR作用后Survivin mRNA的表达,免疫组化(SABC)方法检测DNR、VCR作用后Survivin蛋白的表达。结果DNR诱导白血病细胞凋亡的能力较VCR强(P<0.05),Sur-vivin mRNA和蛋白表达在DNR组明显低于VCR组,呈时间和剂量依赖性,差异具有统计学意义(P<0.05)。结论Survivin在DNR作用机制中发挥重要作用,在VCR作用机制中没有重要作用。
Objective To investigate the role of Survivin in the mechanism of daunorubicin (DNR) and vincristine (VCR). Methods The Molt-4 cells with high expression of Survivin were selected in the pediatric blood laboratory of Tongji Hospital, Wuhan University, in 2005. The apoptosis of Molt-4 cells was detected by flow cytometry. The apoptosis of Molt-4 cells was detected by reverse transcription polymerase chain reaction (RT- PCR) was used to detect the expression of Survivin mRNA after DNR and VCR. The expression of Survivin protein was detected by immunohistochemistry (SABC) after DNR and VCR. Results The DNR induced apoptosis of leukemic cells was stronger than that of VCR (P <0.05). Survivin mRNA and protein expression in DNR group was significantly lower than that of VCR group in a time and dose dependent manner (P <0.05) ). Conclusion Survivin plays an important role in the mechanism of DNR and has no important role in the mechanism of VCR.
