目的:探讨天花粉蛋白(TCS)对卵巢癌细胞株HO8910增殖效应并探讨其机制。方法:采用MTT法检测不同浓度(20、40、80μmol/L)对细胞HO8910增殖的影响;通过流式细胞仪观察细胞(20μmol/L)凋亡过程中细胞周期的变化;Western Blot检测不同浓度(20、40、80μmol/L)72hTCS对HO8910凋亡相关蛋白Bax、Bcl-2的表达。结果:随着TCS浓度的升高,作用时间的延长,TCS对HO8910生长抑制率逐渐增高(P<0.05),呈一定的时间,剂量依赖性。流式细胞仪检测随着天花粉蛋白浓度的增加,Bax基因的表达增加而Bcl-2的表达减少。结论:TCS对卵巢癌细胞HO8910细胞增殖抑制作用,该作用可能与其下调Bcl-2及上调Bax蛋白表达有关。
Objective: To investigate the effect of trichosanthin (TCS) on the proliferation of ovarian cancer cell line HO8910 and explore its mechanism. Methods: The effects of different concentrations (20, 40, 80μmol / L) on the proliferation of HO8910 cells were detected by MTT assay. The changes of cell cycle were observed by flow cytometry (FCM) (20,40,80μmol / L) 72hTCS on HO8910 apoptotic protein Bax, Bcl-2 expression. Results: With the increase of TCS concentration and the prolongation of action time, the inhibitory rate of TCS on HO8910 gradually increased (P <0.05), which showed a certain time-and dose-dependent manner. Flow Cytometry showed that as the concentration of trichosanthin increased, the expression of Bax gene increased and the expression of Bcl-2 decreased. CONCLUSION: TCS can inhibit the proliferation of ovarian cancer cell line HO8910, which may be related to its down-regulation of Bcl-2 and up-regulation of Bax protein expression.
