目的:研究食管鳞癌组织中Survivin mRNA与蛋白表达的相互关系,探讨两者在食管鳞癌发生和发展中的作用。方法:采用原位杂交技术检测30例食管鳞癌及相应的癌旁组织中Survivin mRNA的表达,同时采用免疫组织化学SP法检测相应标本中Sur-vivin蛋白的表达。结果:在食管鳞癌和相应的癌旁组织中Survivin mRNA表达的阳性率分别为66.7%和10.0%,Survivin蛋白表达的阳性率分别为76.7%和16.0%。食管鳞癌组远高于癌旁组,差异有统计学意义,P<0.01。Survivin mRNA与蛋白在食管鳞癌组织中的阳性表达具有一致性(P>0.05),其阳性率高低与性别、年龄、肿瘤大小无关,而与组织学分级、淋巴结转移、TNM分期有关。结论:Sur-vivin基因的异常表达可能对食管鳞癌的发生和发展起重要作用。
Objective: To investigate the relationship between Survivin mRNA and protein expression in esophageal squamous cell carcinoma (ESCC) and to explore the role of both in the occurrence and development of esophageal squamous cell carcinoma. Methods: Survivin mRNA expression was detected by in situ hybridization in 30 esophageal squamous cell carcinomas and corresponding paracancerous tissues, and immunohistochemical SP method was used to detect Survivin protein expression in the corresponding specimens. Results: The positive rates of Survivin mRNA expression in esophageal squamous cell carcinoma and its corresponding paracancerous tissues were 66.7% and 10.0%, respectively. The positive rates of Survivin protein expression were 76.7% and 16.0%, respectively. Esophageal squamous cell carcinoma group was much higher than the adjacent non-cancerous group, the difference was statistically significant, P <0.01. The positive expression of Survivin mRNA and protein in esophageal squamous cell carcinoma was consistent (P> 0.05). The positive rate of Survivin mRNA was not related with sex, age and tumor size, but correlated with histological grade, lymph node metastasis and TNM staging. Conclusion: The abnormal expression of Sur-vivin gene may play an important role in the occurrence and development of esophageal squamous cell carcinoma.
