目的:探讨大剂量通冠胶囊对动脉粥样硬化兔血一氧化氮(NO)、假血友病因子(vWF)水平及血管内膜、中膜厚度的影响。方法:健康雄性新西兰大耳白兔18只,随机分为空白对照组、高脂组、药物组(通冠胶囊组)共3组各6只,通过免疫性内皮损伤合并高脂饲料诱导法建立动脉粥样硬化兔实验动物模型,对照组喂饲普通饲料,高脂组喂饲高脂饲料,药物组在高脂组的基础上每天加喂通冠胶囊15mg/kg,于实验6周末测定血中总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)、NO、vWF,结束后取左髂外动脉约2cm行血管内皮病理学检测,计算血管腔内膜和中膜的厚度。结果:药物组TC、TG、LDL-C、vWF水平及内膜厚度均低于高脂组(P<0.05);中膜厚度低于对照组及高脂组;HDL-C、NO水平均高于对照组及高脂组(P<0.05)。结论:通冠胶囊能够降低内皮损伤后血vWF水平,同时提高血NO水平,减少血管内膜、中膜厚度,从而抑制内膜增生及血管狭窄。
Objective: To investigate the effects of high dose Tongguan Capsule on the levels of blood nitric oxide (NO), von Willebrand factor (vWF), intima and media in atherosclerotic rabbits. Methods: Eighteen healthy male New Zealand white rabbits were randomly divided into blank control group, high fat group and drug group (Tongguan Capsule group), 3 in each group. 6 rabbits were established by immuno-endothelial injury combined with high-fat diet induction Atherosclerosis rabbit experimental animal model, the control group fed normal feed, high-fat group fed high-fat diet, the drug group on the basis of high-fat group fed Tongguan capsule 15mg / kg daily test 6 weeks at the end of the experiment blood (TC), triglyceride (TG), HDL-C, LDL-C, NO, vWF, and the left external iliac artery Endothelial and pathological examination was performed to calculate the thickness of endothelium and tunica media. Results: The levels of TC, TG, LDL-C and vWF in the drug group were lower than those in the hyperlipidemia group (P <0.05). The thickness of the media was lower than that in the control group and the hyperlipidemia group In control group and high-fat group (P <0.05). Conclusion: Tongguan Capsule can reduce the level of blood vWF after endothelial injury, increase the level of blood NO, decrease the thickness of intima and media, and inhibit the proliferation of neointima and vascular stenosis.
