目的:探讨下调microRNA-214(miR-214)表达对人卵巢癌SKOV-3细胞迁移和侵袭的作用及其可能机制。方法:采用脂质体介导法将miR-214抑制剂转染人卵巢癌SKOV-3细胞,Real-time PCR法检测miR-214、核因子-κB(nuclear factor-κB,NF-κB)和尿激酶型纤溶酶原激活剂(uridylyl phosphate adenosine,u PA)基因mRNA的表达,Western blotting法检测细胞NF-κB和u PA蛋白表达。划痕试验检测细胞迁移能力,Transwell小室法检测细胞侵袭能力。结果:经转染miR-214抑制剂后,人卵巢癌SKOV-3细胞miR-214表达降低,细胞迁移和侵袭能力降低。同时NF-κB和u PA mRNA和蛋白表达也降低。结论:下调人卵巢癌SKOV-3细胞miR-214表达可抑制细胞迁移和侵袭能力,下凋NF-κB和u PA基因表达可能是其作用机制。
AIM: To investigate the effect of down-regulating the expression of miR-214 on the migration and invasion of human ovarian cancer cell line SKOV-3 and its possible mechanism. Methods: miR-214 inhibitor was transfected into human ovarian cancer cell line SKOV-3 by liposome-mediated method. The expression of miR-214, NF-κB and NF- The mRNA expression of uPA in uridylyl phosphate adenosine (uPA) was detected by Western blotting. The expression of NF-κB and u PA were detected by Western blotting. Scratch test to detect cell migration ability, cell invasion assay by Transwell chamber method. Results: After transfected with miR-214 inhibitor, the expression of miR-214 in human ovarian cancer SKOV-3 cells decreased and the ability of cell migration and invasion decreased. At the same time, NF-κB and u PA mRNA and protein expression also decreased. Conclusion: The down-regulation of miR-214 expression in human ovarian cancer SKOV-3 cells can inhibit the migration and invasion ability of cells, and the expression of NF-κB and u PA may play a role in its down-regulation.
