目的:观察大鼠骨髓间充质干细胞在子宫内膜组织的迁移分化情况。方法:第2代雄性大鼠骨髓间充质干细胞行PKH26标记,实验组5只雌性大鼠尾静脉注射1×107/mlPKH26标记BMSCs 1 ml,对照组5只雌性大鼠尾静脉注射生理盐水1 ml。6周后切下子宫制成冰冻切片,荧光显微镜下观察标记细胞在子宫内膜组织的分布情况,并进行免疫荧光染色检测标记细胞角蛋白的表达。结果:实验组大鼠子宫内膜组织均可见标记细胞散在分布于子宫内膜组织中,腺上皮中少数标记细胞角蛋白染色阳性。对照组子宫内膜均未见标记细胞。结论:BMSCs可以迁移至子宫内膜,可以向腺上皮细胞分化,参与子宫内膜的增生。
Objective: To observe the migration and differentiation of rat bone marrow mesenchymal stem cells in endometrial tissue. METHODS: The second generation of rat bone marrow-derived mesenchymal stem cells were labeled with PKH26. In the experimental group, 5 female rats were injected with 1 × 107 / ml PKH26 labeled BMSCs into the caudal vein. In the control group, 5 female rats were injected with saline 1 ml. Six weeks later, the uterus was cut into frozen sections, the distribution of labeled cells in the endometrium was observed under a fluorescence microscope, and the expression of cytokeratin was detected by immunofluorescence staining. Results: The labeled cells were scattered in the endometrium in the endometrium of the experimental group, and a few of the labeled cytokeratin positive staining in the glandular epithelium. Control group, no endometrial marker cells. Conclusion: BMSCs can migrate to the endometrium and differentiate into glandular epithelial cells and participate in the proliferation of the endometrium.
