目的:探讨Fbxw7表达对结肠直肠癌氟尿嘧啶(5-FU)耐药性的影响。方法:采用免疫组织化学,检测53例结肠直肠癌病人肿瘤组织Fbxw7的表达,分析Fbxw7表达与术后接受5-FU化疗的肿瘤复发的关系。shRNA转染至结肠癌细胞HCT116,下调Fbxw7后,CCK-8实验和平板克隆形成实验检测5-FU处理后的细胞增殖能力。结果:Fbxw7在复发病人的阳性率为20.0%,未复发病人的阳性率为53.6%。Fbxw7表达与术后肿瘤复发相关。5-FU处理结肠癌细胞后,Fbxw7的mRNA和蛋白质水平均明显下降。与未下调Fbxw7细胞相比,Fbxw7的下调促进5-FU处理后细胞的相对活力和增殖能力。结论:下调Fbxw7增强结肠癌细胞的5-FU耐药性。
Objective: To investigate the effect of Fbxw7 expression on the drug resistance of 5-fluorouracil (5-FU) in colorectal cancer. Methods: Immunohistochemistry was used to detect the expression of Fbxw7 in 53 patients with colorectal cancer. The relationship between Fbxw7 expression and tumor recurrence after 5-FU chemotherapy was analyzed. shRNA transfected into colon cancer cells HCT116, down-regulation of Fbxw7, CCK-8 experiments and plate cloning formation assay 5-FU treatment of cell proliferation. Results: The positive rate of Fbxw7 was 20.0% in patients with recurrent disease and 53.6% in patients without recurrent disease. Fbxw7 expression correlated with postoperative tumor recurrence. 5-FU treatment of colon cancer cells, Fbxw7 mRNA and protein levels were significantly decreased. Down-regulation of Fbxw7 promoted the relative viability and proliferation of cells after 5-FU treatment as compared to non-down-regulated Fbxw7 cells. Conclusion: Down-regulation of Fbxw7 enhances 5-FU resistance of colon cancer cells.
