目的 旨在探讨肌肉注射成纤维细胞生长因子受体(FGFR)活化抑制剂———5’甲基硫代腺苷(5’MTA)对大鼠体内反应性星形胶质化的调控作用。方法 Wistar大鼠20只均分为2大组:甲组损伤后4d处死(含5只加药鼠),乙组损伤后8d处死(含5只加药鼠);两组中非加药鼠作相应单纯损伤对照组。在冰冻和石蜡切片上行HE染色和免疫组织化学染色观测碱性成纤维细胞生长因子(bFGF)和胶质纤维酸性蛋白(GFAP)的表达并作形态定量分析。结果 5’MTA加药组在伤后4d其伤道旁细胞数量以及bFGF和GFAP的反应强度均明显低于对照组;加药组伤道宽度(8d时)明显大于对照组,提示其愈合受到5’MTA阻抑。结论 肌肉注射5’MTA能够部分抑制大鼠反应性星形胶质化
Objective To investigate the regulation of reactive astrogliosis in rats by intramuscular injection of fibroblast growth factor receptor (FGFR) activation inhibitor 5 ’methylthioadenosine (5’MTA). Methods 20 Wistar rats were divided into 2 groups: group A was killed 4 days after injury (including 5 dosing rats), group B was sacrificed 8 days after dying (including 5 dosing rats); two groups of non-dosing rats For the corresponding simple injury control group. The expression of basic fibroblast growth factor (bFGF) and glial fibrillary acidic protein (GFAP) were observed by HE staining and immunohistochemical staining on frozen and paraffin sections, and the morphological quantitative analysis was performed. RESULTS: The number of injured para-cells and the reaction intensity of bFGF and GFAP in the 5-MTA group were significantly lower than those in the control group 4 days after injury. The width of the wound in the drug-added group (8 d) was significantly greater than that of the control group, MTA suppression. Conclusion Intramuscular injection of 5’MTA can partially inhibit reactive astroglization in rats
