目的体外细胞培养观察利胆药物熊脱氧胆酸(ursodeoxycholic acid,UDCA)及其衍生物牛磺熊脱氧胆酸(tauroursdeoxycholic acid,TUDCA)或利福平(rifampicin,Rif)对多耐药相关蛋白3(multidrug resistance-associate protein 3,MRP3)及其核受体FTF(fetoprotein transcription factor)表达的影响。方法用UDCA及其衍生物TUDCA或Rif分别刺激培养的肝癌细胞HepG2后,抽提HepG2的总RNA与细胞总膜蛋白、核蛋白,分别用Quantitative real-time PCR和Westernblot检测MRP3和FTF mRNA与蛋白水平表达的变化。结果UDCA及其衍生物TUDCA与Rif均可诱导HepG2细胞内核受体FTF表达增高,同时MRP3 mRNA和蛋白水平表达也相应增高。结论UDCA及其衍生物TUDCA和Rif刺激肝癌细胞HepG2细胞膜转运蛋白MRP3表达上调可能与核受体FTF途径相关。
OBJECTIVE: To observe the effect of ursodeoxycholic acid (UDCA) and its derivative tauroursdeoxycholic acid (TUDCA) or rifampicin (Rif) on multidrug resistance-related protein 3 (multidrug resistance-associate protein 3, MRP3) and its nuclear receptor FTF (fetoprotein transcription factor) expression. Methods After HepG2 cells were cultured with UDCA and its derivatives TUDCA or Rif, the total RNA and total cellular proteins of HepG2 were extracted. The mRNA and protein of MRP3 and FTF were detected by Quantitative real-time PCR and Western blot, respectively Changes in the level of expression. Results UDCA and its derivatives, TUDCA and Rif, both induced an increase in FTR expression in HepG2 cells and a corresponding increase in MRP3 mRNA and protein expression. Conclusion UDCA and its derivatives TUDCA and Rif stimulate HepG2 cell membrane transporter MRP3 expression may be related to the nuclear receptor FTF pathway.
